This is by far the most lucrative method for earning billions in single-player. Crucially, save Lester's assassination missions until after you've completed the main story. This allows you to invest all three characters' combined wealth into the stock market. Before each assassination mission, Lester will give you a target company. Invest heavily in a rival company (or the target company if Lester advises it will rise after the mission).
