Doku is a formidable samurai demon who appears twice. In Chapter 2, he's unkillable; the fight is meant to be lost. Focus on surviving and learning his patterns. In Chapter 13, you face him for real. He wields a massive katana and has a powerful grab attack that drains your health. His attacks are slow but hit incredibly hard. Block his sword swings and counter-attack. When he charges his grab, dodge immediately. The Flying Swallow is effective for hitting him quickly. Use Ninpo when he's open. After taking enough damage, he'll summon ghost fish; clear them quickly with wide-sweeping attacks or Ninpo. Reward: Dark Dragon Blade (Chapter 13).
