Utilize healing items if necessary. After dispatching the initial wave, a cutscene will introduce the mysterious assassin, who will engage Kiryu in a brief, challenging encounter. You won't defeat him in this first meeting, but focus on surviving and landing some hits. This encounter highlights the increased threat level and the personal stakes involved.
